Cytokinins — shoot proliferation & cell division
Cytokinin
6-Benzylaminopurine (BAP)
CAS: 1214-39-7
The most widely used synthetic cytokinin in plant tissue culture. BAP promotes shoot proliferation, axillary bud development, and adventitious shoot formation across a broad range of plant species. Typically used at 0.1–10 mg/L in MS or B5 medium.
Cytokinin
Kinetin
CAS: 525-79-1
A naturally occurring cytokinin (N⁶-furfuryladenine) that stimulates cell division, shoot initiation, and delays senescence. Often used in combination with auxins to regulate the balance between shoot and root organogenesis in callus culture systems.
Cytokinin (ultra-potent)
Thidiazuron (TDZ)
CAS: 51707-55-2
A phenylurea-type compound with exceptionally potent cytokinin activity — effective at nanomolar concentrations (0.001–1 mg/L). TDZ promotes somatic embryogenesis and shoot organogenesis, particularly effective in woody and recalcitrant plant species that respond poorly to conventional cytokinins.
Auxins — root induction & callus formation
Auxin
Indole-3-butyric acid (IBA)
CAS: 133-32-4
The preferred auxin for adventitious root induction in shoot cultures and microcuttings. IBA is chemically stable in culture media and less prone to degradation than IAA, making it the auxin of choice for rooting protocols. Used at 0.1–5 mg/L depending on species.
Auxin (synthetic)
Naphthalene acetic acid (NAA)
CAS: 86-87-3
A stable synthetic auxin widely used for root induction and callus initiation. NAA is resistant to enzymatic degradation and maintains consistent activity throughout extended culture periods. Also used in somatic embryogenesis protocols and fruit set applications. Used at 0.01–5 mg/L.
Auxin (natural)
Indole-3-acetic acid (IAA)
CAS: 87-51-4
The principal natural auxin in plants — regulating cell elongation, apical dominance, root initiation, and tropisms. In tissue culture, IAA is used for rooting and maintaining callus at low concentrations. Typically used at 0.01–1 mg/L.
Auxin (synthetic · potent)
2,4-Dichlorophenoxyacetic acid (2,4-D)
CAS: 94-75-7
A potent synthetic auxin primarily used for callus induction and somatic embryogenesis. At low concentrations (0.1–2 mg/L), 2,4-D strongly promotes dedifferentiation and embryogenic callus formation. Essential for cereal and many dicot embryogenesis protocols.
Gibberellins — elongation & germination
Gibberellin
Gibberellic acid (GA₃)
CAS: 77-06-5
The most biologically active and commercially important gibberellin. GA₃ promotes stem elongation, seed germination, breaking of dormancy, and fruit development. In tissue culture, GA₃ is used to promote elongation of microshoots, overcome dormancy in recalcitrant seeds, and stimulate germination of somatic embryos. Used at 0.01–1 mg/L in culture medium.
